Research Summary
SS-31 has been evaluated for primary mitochondrial myopathy in the Phase 3 MMPOWER-3 trial (NCT03323749), which was preceded by the MMPOWER Phase 1/2 dose-escalation study (NCT02367014). MMPOWER-3's primary endpoints (6-minute walk test improvement and total fatigue) were not met and the trial was terminated, though long-term extension data showed durable improvements in some completers. A separate Phase 3 trial in nuclear-DNA primary mitochondrial disease (NCT05162768) has since completed. The heterogeneous nature of PMM may complicate endpoint selection.
