Research Summary
Vitiligo is an autoimmune condition causing depigmented patches due to melanocyte destruction. CLINUVEL Pharmaceuticals has investigated afamelanotide as a melanocyte stimulant to repigment affected skin, particularly in combination with narrowband UVB (NB-UVB) phototherapy. Early phase studies showed promising repigmentation rates when afamelanotide was combined with NB-UVB versus NB-UVB alone. A Phase 2 trial (VIPHOT, PMID 28078665) found significantly faster and more complete repigmentation with the combination approach. CLINUVEL initiated Phase 3 development (CUV-105) for vitiligo, though afamelanotide is not yet approved for this indication in any jurisdiction. This is an investigational use only. Any repigmentation benefit would require the patient to undergo light therapy in conjunction with the implant, under medical supervision. The melanocyte-stimulating mechanism is biologically plausible, but regulatory approval is pending.
