Research Summary
The first-in-human NCT03715985 study enrolled 12 patients with metastatic melanoma receiving anti-PD-1 therapy; its primary endpoint was safety. The 2024 paper reported 8 objective responses among 12 patients (six partial, two complete) and vaccine-specific T-cell responses in all 12. The 2022 interim report described the first five participants of this same cohort, not another five-patient trial. The distinct NCT05309421 phase 2 study enrolled 17 checkpoint-inhibitor-naive patients with unresectable or metastatic melanoma and used a 12-week pembrolizumab run-in. A sponsor's AACR 2026 poster reported 12 objective responses among 16 analyzed participants and immune results in 15. The phase 2 primary response-conversion endpoint was not formally analyzed in the 2025 sponsor account. Neither single-arm study proves EVX-01 improves outcomes beyond anti-PD-1 therapy; EVX-01 remains investigational.
